Collagenase B, Animal Component-Free (ACF) is obtained from cultures free of animal-derived materials. Collagenase is a protease consisting of a single polypeptide chain approximately 1,000 amino acid residues in length. Collagenase is capable of digesting native collagen fibrils commonly found in connective tissues and therefore is frequently used for tissue dissociation. Collagenase preparations contain the activity of several proteases, including collagenase, caseinase, clostripain, and trypsin (Kessler & Yaron). Collagenase B, ACF contains higher collagenase and caseinase levels than Collagenase A and is intended for use in applications where it is necessary to prevent the introduction of potential animal-derived pathogens.

Collagenase B is prepared from Clostridium histolyticum cultures by filtration ammonium sulfate precipitation dialysis and lyophilization. Collagenase from Clostridium histolyticum is a collagenolytic enzyme. It interacts with collagen-like substances and is responsible for the degradation of collagen into small peptides. It is widely used for the disaggregation of many types of tissues (e.g. lung heart muscle bone adipose tissue liver kidney cartilage mammary gland placentae blood vessels brain tumors) and for the preparation of single-cell suspensions for the establishment of primary cell culture systems. Collagenase B is recommended when yield and viability are important.

Trypsin is a major proteolytic enzyme synthesized as a preproenzyme by pancreas and is stored as proenzyme trypsinogen in secretory granules. Trypsin belongs to the family of serine proteases that are characterized by the catalytic triad His57 Asp102 and Ser195. Trypsin is routinely used in proteomics research for peptide mapping and protein sequence work due to its highly specific cleavage resulting in a limited number of tryptic peptides. Trypsin is a pancreatic serine endoprotease which hydrolyzes peptide bonds specifically at the carboxyl side of arginine and lysine residues. The rate of hydrolysis is slower if an acidic residue is on either side of the cleavage site and cleavage may not occur if a proline residue is on the carboxyl side.
